I previously exported site profiles and am now trying to import them (all of mine disappeared for some reason). When I follow the instructions and right click on the site profile tree, select import and select the file previously exported, nothing happens. Is there something I am missing, or does the file name or extension need to be specific?

Try selecting the "reindex sites" option.
If that doesn't work, it could potentially be a permissions issue. Exit Core FTP, right click on the Core FTP icon, run as admin, and try again.
There is also the command line option available:
Core FTP install path\coreftp.exe -import <exported_sites_filename>
